Follow-up Comment #2, bug #46826 (project grub): Do you use initrd? Linux kernel replaces "/" in root= argument with "!" when looking it up, so it should be entirely equivalent; it sounds more like your initrd fails to handle it. Could you attach "udevadm info --export-db"?
