Nope.  It just popped into my head. :-)

How and where should the knowledge about default destination directories
be stored?

My idea is to put this in one config file at a standard location which can include site specific information. This could include information such as special compile/link flags required for a particular architecture format and expected subdirectory or library file naming. It would list architecture formats which apply for that system.

Which tools can be safely used to query the desired informations?

I am not aware of any such tools. Certainly test compiles can be done with various flags to see what the compilation tools are capable of, but the result might not be suitable for the current system.
