So the presence of ISO 9660 on the same medium is not the problem.

I am out of guesses. Everything points to something being missing or going
wrong in GRUB.
(I think the theory of intentionally blocking GRUB is fewly plausible
 given the experiment results with SYSLINUX.)

Its boot software differs much between BIOS and EFI. Look at the size of the
EFI System Partition (partition 2, 15 MB). Maybe the EFI programs are
more eager to pull everything from the stick than the BIOS programs are.
Booting from USB stick is available in Knoppix ISO only since 8.0, march 2017.
Earlier versions only booted from DVD. Both versions can be installed to
USB stick after successful boot. This is qualified unpacking, though, not
the same as dd'ing the ISO onto the stick.

You could try on some other machine which offers BIOS legacy mode.
Probably Klaus Knopper is interested in learning where Knoppix looks good
and where it looks bad.
