Is LilyPond installed with administrative rights?  That's a common reason for drag and drop not to work (it's forbidden in that case).

I installed Lilypond pn Windows 10. I followed the intstructions of updating the path. Then I put the test.ly file on the desktop. Then placed it's icon over the Lilypond icon.

Two files were created: test and test-tmp-9972888,

This is the content of test:
